Biography

Endothelial dysfunction could be due to aberrant growth pattern or localized signal perturbations that results in growth malformation, compromised vascular function, increased cardiovascular disease risk, etc. My research is focused on understanding the endothelial cell function / dysfunction in vascular anomalies such as infantile hemangioma, Sturge-Weber syndrome and Cobb syndrome. My lab had been studying the endothelial cell function under moderate and excess levels of oxidative stress and its effects on vascular integrity and physiology. We specifically follow two catabolizing enzymes viz., nitric oxide synthase and indoleamine 2,3-dioxygenase to understand bioavailability of vasodilators and other catabolites that contribute to endothelial function, growth and cell death. Using proteomic, imaging and biochemical tools we are attempting to elucidate patterns of endothelial cell function / dysfunction.

My lab also studies congenital heart disease associated endothelial dysfunction. Genetic mutations that affect vital regulatory and functional gene products cause disease conditions and the manifestation of the disease is depicted in many ways including endothelial dysfunction. My lab studies the fatty acid deficiency related heart disease and are focused on understanding the relationship between the heart condition and endothelial cell function in disease progression.

I also serve as the Scientific Director of Children’s Wisconsin Research Institute's Imaging Core and Фβͼ – Neuroscience Research Institutes Imaging Core and I teach microscopy and analytical methods along with hands on training on several light microscopic instruments and scanners. In this collaborative environment, I advise on experimental design, sample preparation, data acquisition, processing and analyzing data. The core houses bright field, fluorescence, confocal, 2 photon confocal, laser micro dissection, laser scanning cytometry, fluorescence slide scanning and bright field slide scanner. The facility enables acquisition of high quality 2D, 3D, 4D and 5D images and quantification by several methods for reproducible, automatic and high levels of accuracy. My lab collaborates with several of the investigators and participate in their studies to further their knowledge and understanding.
